Two different things glow at night in the Keys. One is bioluminescence: plankton that flash when the water moves, on warm, moonless nights, never on a schedule. The other is the nightly clear-kayak tour, where LED lights in the hull light up the flats beneath you. This page tells them apart, then shows you what to book.
Short answer: No Key West tour sells natural bioluminescence, and none can promise it. The nightly Luminous Lagoon tour paddles LED-lit clear kayaks over the flats: the kayak lights the water, and you watch rays, fish and the bottom go by underneath you. It's $79.95 a kayak and a great night out. Natural glow does show up in the Keys on warm, dark nights, and if it fires while you're out there, that's a bonus. If a guaranteed bioluminescent bay is the goal, that's Puerto Rico.
What you're booking
Transparent hulls with lights built in. The glow under your seat comes from the kayak, and it lights the bottom, seagrass and whatever swims through.
Stingrays lifting off the sand, small nurse sharks, needlefish, crabs. Night is when the flats get busy, and the lit hull is your window.
One departure nightly at 8 PM, about 90 minutes on the water. Guided, with a short paddling briefing first.
Ages 4 and up. Kids ride in a double with an adult. No kayak experience needed; the route is shallow, protected water at an easy pace.
Key West. Singles and doubles; the double is the practical pick for a parent paddling with a younger kid.
Clothes that can get wet, a dry layer for winter nights, and a dry bag for the phone. Phone cameras struggle in the dark; watch with your eyes.

Thirty miles up the Keys on Big Pine. Launch at sunset in clear kayaks, then paddle mangrove tunnels and hidden canals into the dark. Stingrays, baby sharks, turtles and manatees are common. All ages; one paddler 16+ per kayak.

The real thing
Dinoflagellates, single-celled plankton, flash blue-green when the water around them is disturbed. A paddle stroke, a fish, your hand trailing over the side.
Warm water builds the population, so late spring through fall is the season. The week around the new moon is the window: a bright moon washes the glow out the way city lights wash out stars. Overcast nights help.
Shallow backcountry flats and mangrove edges away from the lights of Duval. The darker the water, the more you see.
Honestly, variable. Some nights every stroke draws a streak; most nights it's a faint shimmer, and some nights nothing. No Keys operator sells it as a guaranteed show, and you should be wary of any that does.
Book a night paddle in summer or early fall in the new-moon week, and ask the guide to kill the LEDs for a few minutes once your eyes adjust. You'll see whatever the water is doing that night.

Tips
Darkest water. Check a moon calendar before you lock dates, whether you're hoping for natural glow or just want the LED show to pop.
Your eyes need 20 minutes to adjust, and one phone light resets everyone. Red mode if you need light.
Fingers in the water at paddling speed. If the plankton are up, this is how you'll know.
Every stroke drips on you. Winter nights on the water run about 10 degrees cooler than the dock.
Questions
There is a nightly clear-kayak tour in Key West, but its glow comes from LED lights in the kayaks, not from bioluminescent plankton. Natural bioluminescence does occur in the Keys on warm, moonless nights, but no operator guarantees it.
